Analytical Determination of Superoxide Dismutase Enzyme in Plant Materials Using Ransod Set

The activity of superoxide dismutase (SOD; EC 1.15.1.1) in several barley grain samples was measured using a diagnostic Ransod set, which is utilized for determination of SOD in blood. The determination of SOD activity is based on inhibition of the reaction of xanthine and xanthine oxidase with a tetrazolium salt and measurement of absorbance of the produced formazane at 505 nm and 37 °C. The calibration curve was linear. The SOD activity of a Krona x Kompakt sample was (108.2±1.6) U/g dry matter. A significantly higher antioxidant activity was found in several samples, which are recommended for production of malt or food and fodder. The determination of SOD activity in barley and malt using the Ranson set is simple and reliable.
