Image Analysis and Activity of Intracellular Esterases as New Analytical Tools for Determination of Growth and Viability of Embryonic Cultures of Spruce (Picea sp.) Treated with Cadmium

The influence of Cd-EDTA (50, 250 and 500 μmol l-1) on spruce cultures was investigated by image analysis of early somatic embryos (ESEs) of spruce. Image analysis with double staining can be used for the determination of viability of the culture. The glutathione content was studied by adsorptive transfer stripping differential pulse voltammetry. The cluster area decreased and the number of dead cells increased with increasing concentration of Cd and the time of Cd treatment. The glutathione content was nine times higher in ESEs treated with 500 μmol l-1 Cd-EDTA after twelve days in comparison with the control.
